How to Reconstitute Peptides: A Step-by-Step Research Guide

Proper reconstitution is critical for peptide research. This guide covers bacteriostatic water, dilution math, sterile technique, and storage best practices.

Reconstitution is the process of dissolving a lyophilized (freeze-dried) peptide powder in a liquid solvent to create an injectable solution. Getting this right is fundamental to accurate peptide research.

What You Need

  • ·Lyophilized peptide vial
  • ·Bacteriostatic water (BW) — 0.9% benzyl alcohol preservative
  • ·Alcohol swabs
  • ·Insulin syringes (typically 1 mL / 100 IU)
  • ·Clean, dry work surface

Why Bacteriostatic Water?

Bacteriostatic water contains 0.9% benzyl alcohol, which inhibits bacterial growth and extends the shelf life of reconstituted peptides to approximately 30 days when refrigerated. Sterile water for injection (without benzyl alcohol) can be used but reconstituted solutions should be used more quickly.

The Math

To calculate how much BW to add, use this formula:

Volume of BW (mL) × 100 = total IU capacity
Peptide amount (mcg) ÷ IU per dose = mL per dose

Example: 5 mg (5,000 mcg) vial + 2 mL BW = 2,500 mcg per mL. A 250 mcg dose = 0.1 mL = 10 IU on an insulin syringe.

Step-by-Step Process

1. Allow the peptide vial and BW vial to reach room temperature

2. Wipe the rubber stopper on both vials with a fresh alcohol swab

3. Draw the desired volume of BW into the syringe

4. Insert the needle into the peptide vial at an angle, allowing BW to run down the side of the vial — do not squirt directly onto the powder

5. Swirl gently (do not shake) until fully dissolved

6. Label the vial with the date of reconstitution

Storage After Reconstitution
